Phosphoglycerate mutase (PGM) enzymes catalyse the isomerisation of phosphoglycerate substrates, a process essential for the metabolism of glucose and/or 2,3-phosphoglycerate in nearly all organisms. This clan brings together all such enzymes, except the 2,3-diphosphoglyceric acid independent PGMs, which are unrelated [1]. The catalysis of all of the PGMs in this clan proceeds via a phosphohistidine-enzyme intermediate, which re-orientates the substrate in the catalysis process. The more divergent fructose-2,6-biphosphatase and acid phosphatases are also included, due to their structural and functional similarity. All of the above enzymes phosphorylate a histidine residue during their catalytic activity which requires two main reactions, phosphatase and phosphotransferase. Some of these enzymes specialise in just one reaction, the phosphatase reaction, but all have the ability to perform both of them albeit at different rates. All of these enzymes are evolutionarily, adopting a Rossmann-like fold at the core of the enzyme, which has become elaborated over time. The differences in this elaboration adjacent to the active site presumably resulted in the catalytic differences leading to different enzymes [1].
